This indicates the protective effect of polyhydroxylated fullerene. Results from the work of Milic Torres et al. confirm the hypothesis and complete previous research showing that fullerenol has high antioxidative and cytoprotective potential without recorded side effects. Considering the mechanisms of DOX toxicity (mainly due to free radical production), prior research and our experimental results conclude that fullerenol’s protective role is based on its high antioxidant potential. Due to its unique electrochemical features, fullerenol acts as a free radical sponge and/or removes free iron by forming a fullerenol-iron complex, preventing further damage to cells by reactive oxygen species (ROS).

A possible reaction mechanism for the formation of fullerenol with a high number of hydroxyl groups involves the hydroxide ion (–OH) inducing hydroperoxide ion (–OOH) formation due to the slightly higher acidity of H2O2 compared to H2O. The formed –OOH attacks C60 to give fullerene epoxide C60O, followed by –OH attack and protonation. The obtained fullerene epoxide is susceptible to further nucleophilic attacks by –OH and –OOH because of its higher strain.
